발표주제: Expand your understanding of biology with Illumina single-cell and spatial sequencing

Single-cell sequencing examines the genomes, epigenetics, transcriptomes, or proteomes of individual cells, and provides an unbiased high-resolution view of cell-to-cell variation. Highly sensitive single-cell RNA sequencing (scRNA-Seq) and multi-omics methods enable assessment of the individual contribution of single cells among complex tissues by profiling their transcriptomes – providing an ideal method to understand heterogeneous subpopulation responses to treatments or environmental cues. Further, these assays enhance the study of cell function and heterogeneity in processes such as differentiation, proliferation, and tumorigenesis. In combination with spatial transcriptomics, the researchers can address the tissue location where transcriptomic profiles and interactions of special cell subpopulations occur. Join us to learn about the recent advances that are enabling high throughput analysis while examining hundreds to tens of thousands of cells per experiment in a cost-effective manner. 발표주제: Single-Cell Library prep, A robust microwell-based single-cell partitioning system for high-dimensional biology research

Since single-cell RNA sequencing technology improved in terms of throughput, sequencing cost per cell, and qualities, it is getting popular throughout various researches. Most single-cell RNA sequencing platforms utilize an unbiased sequencing approach. While powerful, it typically delivers shallow sequencing data for every single cell. To understand the complex heterogeneity of biology, single-cell analysis needs to be designed more elaborately with deeper experimental considerations. One of the strategic approaches is to sort out phenotypic populations before single-cell analysis. With that, it can give a better resolution to define rare subpopulations that might conduct significant roles in complex heterogeneous biology, although the no. of analyzed cells wasn't increased much.

발표주제: PBMC single-cell transcriptomics reveals immunoregulation of COVID-19 during disease progress and corticosteroid treatment

The COVID-19 is a mild to moderate respiratory tract infection in the majority, but can cause life-threatening respiratory failure or persistent debilitating symptoms in a subset of patients. However, the mechanism of protective immunity in mild cases and the pathogenesis of severe COVID-19 remain unclear. We performed single cell transcriptome analysis of peripheral-blood mononuclear cells in a three-cohort study, in total 171 clinical patient samples. We identified molecular signature of hyperinflammation in severe COVID-19, different sets of cytokines and growth factors associated with disease aggravating or improving processes as well as immune-regulatory mechanisms induced by dexamethasone treatment. Our study demonstrates detailed insights into the immunopathogenesis of SARS-CoV-2 infection and reveals molecular and cellular characteristics of peripheral immune system during disease progress and treatment of COVID-19 patients.
